Fig. S1. Expression of Rac3 does not affect the activation of endogenous Rac1. Rac activity assay was performed in ev (pcDNA3.1) or Rac3 expressing cells and the GTP-bound active Rac samples (middle panel) were compared to total lysates of the same transfectants (upper panel).
Fig. S2. Constitutively active Rac1 and Rac3 mutants induce similar morphological changes, in contrast to their wild-type counterparts. Cells were transfected with Rac1G12V and Rac3 G12V (together with 5:1 eGFP). After 24 hours, the cells were fixed and stained with phalloidin to visualize filamentous actin. Note that both constitutively active mutants induce a similar morphology including cell spreading; note also the formation of peripheral ruffles at the lamellipodia.
