  Fig. S1. Expression of Rac3 does not affect the activation of endogenous Rac1. Rac activity assay was performed in ev (pcDNA3.1) or Rac3 expressing cells and the GTP-bound active Rac samples (middle panel) were compared to total lysates of the same transfectants (upper panel).

• Supplemental Figure 2 -

  Fig. S2. Constitutively active Rac1 and Rac3 mutants induce similar morphological changes, in contrast to their wild-type counterparts. Cells were transfected with Rac1^G12V and Rac3 ^G12V (together with 5:1 eGFP). After 24 hours, the cells were fixed and stained with phalloidin to visualize filamentous actin. Note that both constitutively active mutants induce a similar morphology including cell spreading; note also the formation of peripheral ruffles at the lamellipodia.
