Fig. S1. The srv2-201 allele, which disrupts profilin binding, results in increased cell size compared with wild-type cells. Isogenic yeast strains, SRV2, srv2-201 and srv2Δ, were grown to log phase at 25°C, then DIC images of cells were captured. The areas of unbudded cells and mother cell compartments of budded cells were measured using OpenLab software and graphed using SigmaPlot. The data show that srv2-201 cells have an increased average cell area compared with wild-type cells (32 μm2 and 25 μm2, respectively, P<10−10), and that the srv2Δ mutation causes an even more severe increase in average cell size (65 μm2).
