Movie 1. 3D image of Cdc8 colocalising with actin in fission yeast. Reconstruction of deconvolved 0.2 μm z-series images of fixed S. pombe cells co-stained with Rhodamine-phalloidin (red) and Cdc8 antisera (green). The proteins colocalise to filaments throughout the cell cycle.
Movie 2. 4D time-lapse series of Cdc8 filament dynamics in vivo. Four-dimensional time-lapse movie of DM809 cells. Each frame is separated by 0.2 seconds, and contains a stack projection of a series of 20 z layers, 0.2 μm apart. GFP-Cdc8 fluorescence reveals the dynamic nature of actin filaments in vivo, and illustrates how these are incorporated into the CAR during mitosis.
