  Fig. S1. Fibrillin-1 can interact with fibronectin. (A) Human fibrillin-1 and FN fragments used in solid-phase binding assays. (i) Schematic diagram showing the fibrillin-1 and FN recombinant fragments, and the FN regions found to interact with fibrillin-1. (ii) Fibronectin fragments used in solid phase binding assays. Lane 1, FN 30 kDa fragment (Sigma); Lane 2, FN 50 kDa fragment (from M. J. Humphries, Manchester, UK); Lane 3, FN H120 fragment (residues 1631-1991, with N-glycosylation) (from M. J. Humphries, Manchester, UK). Fragments were run under reducing conditions on a 4-12% Bis-Tris gel in MES buffer, and stained with Colloidal Coomassie Blue stain. (B) Interactions between recombinant fragments identified using solid-phase binding assays. (i) Fibrillin-1 fragments PF5 and PF7 bind the FN 30 kDa N-terminal fragment; (ii) fibrillin-1 fragment PF5 binds the FN 120 kDa fragment; (iii) fibrillin-1 N-terminal fragments PF1 and PF2 bind the FN 50 kDa cell-binding fragment. These experiments were repeated three times with similar results.
