  Fig. S1. Strains UMI129 Pcrg1:cdc24-GFP and UMI130 bem1nar1 Pcrg1:cdc24-GFP were grown in minimal medium with glucose (non-repressing conditions for Pnar1 and repressive conditions for Pcrg1) and then shifted to rich medium with arabinose (repressive conditions for Pnar1 and inductive conditions for Pcrg1) for 6 hours. UMI131 cdk5ts Pcrg1:cdc24-GFP cells were grown in rich medium with glucose at 22°C (repressive conditions for Pcrg1 and permissive conditions for cdk5ts) and then shifted to rich medium with arabinose at 34°C for 6 hours (inductive conditions for Pcrg1 and restrictive conditions for cdk5ts). Overexpression of cdc24-GFP results in Cdc24-GFP accumulation at the cell tip, regardless of Bem1 and Cdk5 function.
