The data set from each experiment is represented as a separate box. The boundary of the box is determined by the 25th and 75th percentiles, and the whiskers are determined by the 5th and 95th percentiles. The small red square within each box indicates the value of the mean for each set of data (n=20-50 per set; every D is calculated using a set of six to ten measurements of 10 seconds each). Data was collected with a band-pass emission filter (BP505-550) with excitation at 488 nm. For stimulated cells, data was recorded for 8 minutes post-stimulation from different cells and then averaged. All measurements were performed at 25°C.
