This creates binding sites for 14-3-3 adaptor proteins, whereby DLC1 RhoGAP activity is inhibited, most likely by cytosolic sequestration. 14-3-3 binding furthermore masks a nuclear localization signal spanning residues 423-429, thus preventing DLC1 nucleocytoplasmic shuttling.
