Fig. S1. Nucleolar disorganization is correlated with lamin B1 depletion. HeLa cells were co-transfected with vectors expressing YFP-tubulin and lamin B1-RNAi (LB1-RNAi, for lamin B1 knockdown), as well as with a lamin B1 rescue vector (LB1-RNAi + rescue). At 48 hours post-transfection, cells were fixed, immunoprocessed for fibrillarin detection (red labeling) and observed by confocal microscopy (A). Scale bars: 5 µm. The proportion of cells with a normal nucleolar phenotype (top panel) versus abnormal phenotypes (peri-nucleolar, distorted or diffuse; bottom three panels) was counted in each batch and displayed in a plot (B). As observed previously, the nucleoli of LB1-depleted cells are mainly aberrant (black bar). However, the LB1-depleted cells expressing the rescue vector mostly conserve nucleoli with a normal configuration (gray bar).
