Fig. S1. Characterization of rabbit polyclonal antibodies specific to the novel C terminus of the D2SV protein. E14.5 embryonic ventricular myocyte cultures were transfected with the expression constructs indicated, processed for immunostaining with antibodies specific for D2SV (A,E,I,M) and myc (B,F,J,N). Signals were visualized with Alexa Fluor 488 or 555 secondary antibody conjugates. Note the presence of protein aggregates in cultures transfected with a D2SV construct lacking the myc epitope (A). To determine the specificity of staining, transfected cultures were also incubated with D2SV antibodies preincubated with the peptide used for generation of antibodies (I-L). Note the absence of D2SV staining in a cell transfected with the D2SVmyc construct (I). To determine the cross-reactivity of D2SV antibodies with cycD2, cultures transfected with cycD2myc were co-stained with D2SV and myc antibodies (M-P).
