Fig. S1. Coilin RNAi reduces both endogenous coilin and exogenously expressed GFP-tagged coilin. HeLa cells were treated with control (denoted by minus, lanes 1 and 3) or coilin siRNA (denoted by plus, lanes 2 and 4) for 2 days followed by transfection with GFP-WT or GFP-ON coilin (labeled as GC-WT or GC-ON, respectively) for 24 hours. Western blotting for coilin and GFP-tagged coilin was conducted as described in the Materials and Methods. The same blot was also probed with antibodies against tubulin to serve as a loading control. Note that the level of GFP-WT and GFP-ON in coilin siRNA treated cells (lanes 2 and 4) is approximately equal to that of endogenous coilin in control treated cells (lanes 1 and 3). The level of endogenous coilin in coilin siRNA treated cells (lanes 2 and 4) if drastically reduced compared to control siRNA treated cells (lanes 1 and 3).
Fig. S2. The average FRAP recovery curve for each of the GFP coilin mutants. Curves obtained from individual cells were averaged and fitted to the double exponential equation of Kimura and Cook (Kimura and Cook, 2001) as described in the Materials and Methods.
