Fig. S1. Histogram of maximum fluorescence recovery for all Cx43 GJ-like clusters in BICR-M1Rk cells (n=61) as established when fluorescence recovery reached a plateau. Note that 22 GJ-like clusters revealed only 10% maximum recovery, whereas all but two GJ-like clusters exceeded 15% recovery, suggestive of Cx43 within a less-assembled state.
Movie 1. Pleiomorphic vesicles carrying GFP-Cx43Δ2 were observed to reach the cell surface in non-GJs regions and generate bright fluorescent sites. Rapid time-lapse imaging was performed on BICR-M1Rk cells to examine Cx43 delivery to cell-surface domains. Time-lapse fluorescence images and corresponding DIC time-lapse images were superimposed. Movie represents 500 frames with individual scan times of 1.75 seconds. Elapsed imaging time was ∼15 minutes.
Movie 2. Cx43 localizes to membrane protrusions. Time-lapse GFP-Cx43Δ2 fluorescence images were superimposed over synchronous time-lapse DIC images. Movie represents 600 frames with individual scan times of 1.97 seconds. Total imaging time was ∼20 minutes.
