  Fig. S1. Inhibition of IGF-I-mediated phosporylation of Akt and Erk proteins. Results are shown of experiments in which C3H10T1/2 cells were incubated for 30 minutes in serum-free medium with IGF-I 10 nM without or with the PI3-kinase inhibitor, LY294002 (LY) (20 μM), or the MEK inhibitor, UO126 (UO) (10 μM), as described in the Materials and methods. Immunoblots are shown of whole-cell protein lysates for Akt phosphorylated at Ser473 (pAkt^S473), total Akt, tyrosine and serine-phosphorylated Erk1 and Erk2 (pErk1/2) and total Erk proteins.

  Fig. S2. Production of recombinant BMP2. C3H10T1/2 cells were infected with Ad-BMP2, and conditioned medium (CM) was collected as described in the Materials and methods. A representative immunoblot is shown measuring the amount of BMP2 in two aliquots from different batches of CM, with results compared with a standard curve of purified BMP2 purchased from R&D Systems.
