Fig. S1. Expression pattern of cgt-1 and cgt-3 in adult animals. CGT-1::GFP (A-C,G-I) was strongly expressed in the PIV, the excretory system (including excretory cell, duct and canal) and in the pharyngeal muscles cells of the terminal bulb (pm6, pm7 and pm8) (A-C). The expression in the intestinal cells faded by the L3-L4 stages and was no longer visible in adult animals, whereas the IRV and RGC were still very bright during development and in adults (G-I). CGT-3::GFP (D-E,J-L) showed a strong signal in PIV and in the pharyngeal muscles cells of the procorpus (pm3, mc1), the anterior bulb (pm4) and the isthmus (pm5, mc2) (D-E). cgt-3 was still expressed in the intestinal cells in adult animals but more faintly than in larvae, whereas it was still strongly expressed in the IRV and RGC cells (J-L). Intestinal expression was very strong at the L1 stage. Some fluorescence was also visible in a few amphid neurons and in unidentified head cells (not shown). Scale bars: 10 µm.
Fig. S2. The canal formed by the PIV cells and by the IRV and RCG cells is normal in cgt triple mutants. The PIV canal (A) and the IRV and RCG canal (B) were visualized in L1 arrested progeny of LZ196 mothers (which express F54D5.1p::gfp), exposed to cgt-3 siRNAi for 3 days. Z-stacks of PIV and IRV/RGC were created by confocal microscopy and used to reconstitute the canal in three dimensions. Scale bar: 2 µm. (C) The area of the canal section (yellow dashed lines) was similar in L1 cgt-1(qa1809); cgt-2(tm1192) mutants exposed (mut) or not (WT) to cgt-3 siRNAi.
