Fig. S1. A light-microscopic comparison of advanced-age heterozygous and homozygous KO mice. Retina morphology at advanced age WT and Cngb1 KO mice. Even at advanced age heterozygous KO mice (+/−) were normal in appearance throughout all retinal layers. Within 1 year in homozygous KO mice the entire photoreceptor layer is absent; the INL is disrupted and is directly apposed to the RPE.
Fig. S2. QPCR analysis of transcripts encoding retina-specific proteins. Table 1 contains a comparison of body weights of KO and WT mice. Quantitative PCR of ROS gene product transcripts. Relative levels of transcripts encoding several rod photoreceptor-specific proteins were compared in WT and KO retinas. At least three triplicate replicates were done and the data are shown with error represented in SD (n=9-14). Considering the error in the measurements all transcripts were within 25% of WT values except Rpgrip1 that is just out of this range. The data indicate that the changes observed by western analysis for Cnga1, ABCA4 and GC1 are post-transcriptional as none of the genes examined is significantly altered at the transcriptional level.
