Fig. S1. Family-wide analysis of Rab proteins that are specifically localized in the distal portion of the neurites of NGF-differentiated PC12 cells. (A) GFP-Rab3A, -Rab27A, -Rab33A and -Rab37 are mainly targeted to the distal portion of the neurites of NGF-differentiated PC12 cells. pEGFP-C1-Rab3A, -Rab27A, -Rab33A or -Rab37 were transfected into NGF-differentiated PC12 cells, and the cells were examined for GFP fluorescence with a confocal microscope. Note that GFP-Rab3A, -Rab27A and -Rab37 exclusively localized in the neuritis, and that GFP-Rab33A was predominantly present in the neurites as well as in the Golgi-like peri-nuclear region. NGF-differentiated PC12 cells. The localization of GFP-Rabs was classified as being
in the peri-nuclear region (e.g. Golgi, trans-Golgi network, and endosomes), cytoplasm, plasma membrane, and neurites,
where dense-core vesicles are accumulated (Fukuda et al., 2002). Significant
GFP-Rab19 signals were observed in the neurites of PC12 cells, but they did not
colocalize with endogenous Rab27A. *, because of the very low expression level
of GFP-Rab22A we were unable to determine the exact localization of GFP-Rab22A
protein (peri-nuclear or cytoplasm) in PC12 cells.
