Fig. S1. The PI3K inhibitor LY294002 has no significant effect on the organization of the actin cytoskeleton and focal adhesion numbers in HelaS3 cells. (A) Fluorescence micrographs of LY294002-treated cells and control cells. HelaS3 cells were plated on laminin-coated coverslips for 24 hours, treated with either DMSO or 4 μM of LY294002 for another 4 hours and processed for immunofluorescence as described in Fig. 2A. (B) Quantification of focal adhesion numbers was performed as in Fig. 2D.
Fig. S2. Wrch-1 depletion does not affect cell proliferation. Cells were transfected with siRNA targeting luciferase as control (e) or Wrch-1 (h and s for oligonucleotides W-1 and W-2, respectively). Three days after transfection, cells were plated into 96-well plates in the presence of 1% serum. Cell growth was quantified using sulforhodamine B staining. Shown are the averages of six wells (± s.e.m.). Data shown are representative of two independent experiments.
