Fig. S1. Fibronectin interactions with heparan sulfate chains on syndecan-4 trigger signalling, leading to focal adhesion and stress fibre formation, when integrin α5β1 is engaged (Woods et al., 2000; Couchman 2003). We investigated whether MEF syndecan-4 null cells mediated PF9 or PF10 induced stress fibre and focal adhesion formation, when plated on PF8 which allows the cells to engage α5β1 integrin. The syndecan-4 null cells exhibited an unusual rounded morphology, confirming the importance of syndecan-4 in cytoskeletal organisation. There were no focal adhesions in the absence or presence of PF9, the R1691A/R1692A PF9 mutant that cannot bind heparan sulphate, or PF10, although there were prominent circumferential microfilament bundles.
