  • Movie 1 -

Movie 1. CytoD. Treatment with Cytochalasin D and Latrunculin B on BAECs expressing spectrin-cpstFRET (Spec panel), spectrin-C-cpstFRET (Spec-C panel), free cpstFRET (cpstFRET panel) and cpCerulean and cpVenus 1:1 mixture (cpC+cpV panel). Each cell type was presented with FRET ratio image and YFP (Venus) channel image.

  • Movie 2 -

Movie 2. Colchicine. Treatment with Colchicine on BAECs expressing spectrin-cpstFRET (Spec panel) and spectrin-C-cpstFRET (Spec-C panel). Each cell type was presented with FRET ratio image and YFP (Venus) channel image.

  • Movie 3 -

Movie 3. Cell migration. BAECs expressing spectrin-cpstFRET (Spec panel), spectrin-C-cpstFRET (Spec-C panel) and free cpstFRET (cpstFRET panel). Video shows FRET ratio and YFP (Venus) channel for each cell type.

  • Movie 4 -

Movie 4. Cell migration. BAEC expressing spectrin-cptFRET showed increasing stress in spectrin. We observed some cells gradually increased the stress in spectrin during the 6-hour recording period. The cell in this video first elongated the cell body then accelerated the migrating speed then moved laterally to the right. FRET ratio increased from 0.10 to 0.15, suggesting ∼50% more average stress in the spectrin network.
